You bet, the speedo cable unscrews by the knurled collar where it mounts onto the engine.
You may need to use pliers with cardboard "liners" to first loosen it, then undo it by hand. Gently lift straight up when the collar is undone.

You may, by this time, have found that to remove the tank you really should drain the gas and remove the petcock by undoing the two screws which hold it onto the tank underside. Supposedly you're able to remove the tank without removing the petcock, but I've never been able to do it. The petcock either ends up being "forced" over the frame tube, or it scratches the hell out of it. On my bike, there is no way to remove the tank without undoing it. Easy to do, just another step to mess with.

I would definitely remove the tank to paint it. You'll have much more control and if you're going to all the trouble to paint you might as well make it easier to get a good finish. What color are you going to paint it ??
